St. Vincent's Hospital opened in November 1849 in a rented house on 13th Street in Manhattan. Ellen Hughes, Irish-born sister of Archbishop John Hughes, known within her community as Sister Angela, was the hospital's first administrator. Patient rosters at St. Vincent's between 1888 until 1894 indicate a predominantly immigrant, Irish population. By the turn of the century, the patient population majority was native-born.
